CONFOCHECK

Protein secondary structure in aqueous solution in about 30 seconds using a 7 µm AquaSpec flow cell.

Bruker CONFOCHECK measures aqueous proteins with below 0.1 mg/mL detection and 0–95 °C BioATR II temperature control.

CONFOCHECK is a dedicated Bruker FT-IR system for proteins in aqueous solutions. Water absorbs across the amide region used to determine secondary structure; the approximately 7 µm optical path of the AquaSpec flow cell allows the amide bands to be measured through water.

The system measures protein concentration, α-helices and β-sheets, conformational dynamics, thermal unfolding, aggregation, precipitation and crystallization. It also analyzes dissolved buffers, excipients and detergents.

BioATR II uses a silicon micro-ATR crystal with 0–95 °C temperature control, allowing continuous spectra during heating to locate denaturation. Each measurement takes about 30 seconds, with protein-concentration detection below 0.1 mg/mL. Stated service lives are ten years for laser and interferometer and five for the mid-IR source.
