Electron fragmentation at PASEF speed: structural characterisation of glycopeptides and cyclic peptides without sacrificing proteomics throughput.

timsOmni Σ is an ion mobility mass spectrometer with PASEF-EXD electron fragmentation for multiomics, glycopeptide and cyclic peptide analysis.

timsOmni Σ extends the timsTOF Ultra 2 platform by combining ion mobility separation with multimodal electron fragmentation in PASEF-EXD mode. While timsOmni focuses on deep proteoform sequencing, this model targets high-sensitivity multiomics and enhanced bottom-up proteomics.

Electron fragmentation is particularly valuable for compounds that do not respond well to collision-induced dissociation. These include glycopeptides, where the glycan should remain attached to the peptide, and cyclic peptides, which require ring opening. Available modes include ECD, hECD, EID, niECD, EDD and EED, alongside collision-based CCID and RCID. Precursor utilisation in trapped eXd mode exceeds 90%.

The instrument works with CaptiveSpray, VIP-HESI with integrated APCI and Apollo II sources. The manufacturer states that the NEOS source is not compatible with this model.
