Secondary-laser postionisation increases sensitivity by orders of magnitude for lipids and small molecules in tissue imaging.

timsTOF fleX MALDI-2 adds secondary-laser postionisation to improve MALDI imaging sensitivity by one to three orders of magnitude.

timsTOF fleX MALDI-2 adds a secondary postionisation laser to the timsTOF fleX platform. In conventional MALDI, some desorbed material remains neutral and is not detected. The second laser ionises these neutral molecules in the gas phase.

According to the manufacturer, sensitivity increases by one to three orders of magnitude depending on the sample, analyte and matrix. The effect is particularly strong for lipids and certain small molecules that ionise poorly with conventional MALDI. Combined with microGRID, the system supports imaging below 10 µm spatial resolution, including subcellular imaging and single-cell analysis.

Switching between MALDI and MALDI-2 modes is controlled in software, with no hardware changes required.
